The basic building blocks of both DNA and mRNA are adenine, thymine, guanine, and cytosine. These are defined as nitrogen bases and are usually labeled A, T, G, and C. These bases code for twenty specific amino acids. The twenty amino acids that can be created are as follows: alanine, arginine, asparagine, aspartate, cysteine, glutamine, glutamate, glycine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, praline, serine, threonine, tryptophan, tyrosine, and valine. A nucleotide is defined as a linked bunch of molecules, composed of a phosphate group, nitrogenous bases, and a pentose sugar. The nucleotides code for these amino acids in groups of three, giving 64 possible combinations. The groups of threes are called codons. There are actually three codons that don’t code for amino acids. These codons are called stop codons and they signal signal translation termination. These codons are UAA, UAG, and UGA.
The genetic code is said to be degenerate. This means that the code doesn’t code to its capacity. Basically, the twenty known amino acids that can be created by the genetic code can be made by more than one type of nucleotide sequence. For instance, CUU, CUC, CUA, and CUG all code for the amino acid leucine. Another example is that CGU, CGC, CGA, and CGG all code for arginine.
According to the book, there are three main rules for the genetic code. The first rule is that the sequence of nitrogenous bases must follow the direction of translation. mRNA is translated in the 5′ to 3′ direction, so the codon sequences have to occur in the same orientation. This ensures that they will be properly translated. The first base of a codon must be located at the 5′-most end of the codon. For instance, the following three bases code for the amino acid cysteine: CGU. Two codons code for this amino acid, 5′-UGU-3′ and 5′-UGC-3′. 5′-UGC-3′ matches the codon CGU if you read it backwards.
The second rule is that one nucleotide can be used per reading frame. In other words, one nucleotide can only be part of one codon. For example, the code AATT could be read only as AAT or ATT, but not at the same time.
The last rule deals with start and stop codons. Basically, once you begin reading a codon from a specific nucleotide, one must continue reading it by threes until the end. The most common start codon is AUG. UAA, UAG, or UGA are the stop codons. So, the implication of this rule is that any sequence can be read in three different ways, depending on which nucleotide is put first. The three different ways of reading can yield three different amino acids.
Mutations are errors in codons caused by changes in nucleotide bases. Depending on the type of mutation, the error can cause no change or devastating change in protein created. These changes can result in positive phenotypic changes in the living organism, but usually are deleterious. The first type of mutation discussed is the base substitution. This is when one base is substituted for another. There are three main base substitutions: silent mutations, missense mutations, and nonsense mutations. Silent mutations do not change the amino acid created, due to the degeneracy of the genetic code. For example, if UGU is changed to UGC, the corresponding amino acid will still be cysteine. A missense mutation results in a substitution that changes the actual amino acid that is created by the codon. A nonsense mutation is a substitution that actually transforms the codon into a stop codon. This is generally considered to be the worst sort of base substitution mutation because it can really mess up the formation of a protein.
The next category of mutations is the insertion and deletion mutations. The main mutation that correlates to the insertion or deletion of a nucleotide in a codon is the frameshift mutation. This mutation is very profound in its effects, due to it changing every codon in a genetic sequence. It messes up the three codon structure which constitutes code, thus altering the protein that is created by the code.
The last category of mutations are the suppressor mutations. Supressor mutations change the result of an entirely different mutation. There are two types of these: extragenic and intragenic mutations. Extragenic mutations occur outside the genetic code, but has an effect on the amino acid sequence that is translated from the genetic code. Basically, one mutation can negate another mutation, due to the affects of a tRNA mutation. An intragenic mutation comes from within the genetic code. An example of this would be if an insertion of a particular nucleotide was negated by a frameshift deletion of this nucleotide. This book does a thorough job of educating the reader in the basic of the genetic code. It not only defines what the genetic code is, what rules govern it, and what mutations can occur in it, but it discusses how the genetic code is an indicator of evolution. Since all life shares the same four nitrogenous bases, which make up the codons that code for the proteins that make all living bodies, evolution from common descent seems plausible.

For the uninitiated, Buffy Summers was the young girl chosen from a long line of mystical maidens to fight the demons of the dark. She fights vampires with mad martial arts skills with the help of her best friends Willow, a witch and Xander a nice enough ordinary guy. The comic series is well drawn and is surprisingly consistent in its look and feel from issue to issue. The comic book series contains several story arcs but basically it’s Summers and her worldwide army of young slayers against vengeful demons with ulterior motives as well as the full force of the United States Army.
For five seasons Buffy the Vampire Slayer kept us entertained on a fledgling network called the WB then made the jump to UPN for two more seasons before going out with a bang. To the surprise and delight of many of its fans, the “Chosen One” returned in comic book form to continue slaying vampires and demons while taking on the establishment as well. The comic series was called Buffy the Vampire Slayer Season 8 and picked up after the destruction of the Hellmouth and the city of Sunnydale.
Joss Whedon, the creator continued to guide the series in this new format; working feverishly to keep the flavor and style that made the series so unique. The comic series blasted off with Whedon scripting the story and artist Georges Jeanty on pencils and Andy Owens working inks; Dave Stewart created the exquisite colors with Comicraft and Richard Starkings filling out the team on lettering.
Series 7 ended with the group driving off into the great unknown apparently ready to assemble the hundreds of new slayers that have been awakened after the last great confrontation with evil. What we see in the opening segment is a much more emotionally mature Buffy Summers. Our pretty blonde cheerleader has now morphed into General Patton or more appropriately Nick Fury agent of Shield. Her new crew comprising of hundreds of new slayers are battle tested and dedicated to her cause. The TV show was unconventional in its delivery and dialogue and the comic series tries to deliver that same flavor.

If it is a good idea is to you and Vampire Slayer season eight continues many of the relationships that have blossomed throughout the television series while continuing to explore new themes. Even Buffy gets a new love interest that assists in the mending of her broken heart. The mythology that colors the world of Buffy Summers continues to evolve in the color comics. The material continues to feel fresh after all these years because Whedon and the original writers for the show treat the new material with care and pull the reader in with a mix of new characters and old favorites.
Buffy Season 8 started out as an exhilarating new opportunity for comics to continue the storyline and characters past their TV incarnations. Comics can be just as entertaining medium without the exorbitant expense of production. Continuing a TV series in this medium also allows for more merchandising opportunities in new markets. Although the concept of portraying television characters in comics is not new the idea of continuing a series while bridging the gap between television and comics in this way is novel and refreshing.
In Buffy the Vampire Slayer: Season 8, Buffy and her new charges known as Slayers live and train tirelessly in Scotland. The US Army and a roundtable of old and new villains take on Buffy and her friends. A new villain named Twilight figures heavily in the plots and subplots which pepper the series. The slayer army and Buffy are forced to fight for their lives and in defense of their loved ones; as usual, not everyone makes it out alive in a Joss Whedon production. If you are a fan of the show and you are into comics I’m fairly certain you will like this series. Since there are no limitations when it comes to special effects in comics Whedon and the gang have a great time constructing their fantasy world. Although the season 8 story arc is over, Whedon and company are already talking about continuing the project which means lots more work for the Slayer.

]]>“It’s not a grammar book”, Ms Truss claims in the first chapter and proceeds to explain: “The trouble with grammar books is that they are read by keen foreigners; meanwhile, native English-speakers who require their help are the last people who will make the effort to buy and read them.” Ironically, this review of her own book is written by yet another keen foreigner.
“Eats, Shoots & Leaves” is one of the funniest books I’ve ever read. It’s full of jokes; true, but funny, stories; historical examples of how punctuation actually changed people’s lives in the past and genuine examples of misplaced punctuation marks accompanied by hilarious comments. Here and there a short funny poem or a quotation from a famous book saves the reader from being bored (”A cat has claws at the end of its paws. A comma’s a pause at the end of a clause.”).
All of it makes the book a very interesting read. Another impressive thing about the book is the passion, with which the author speaks about the main subject - English punctuation. The main purpose of the book is not to teach people to punctuate correctly (though the book does it), but to attract everyone’s attention to the deteriorating common usage of apostrophes, commas and semicolons. Lynne is doing her best to save English punctuation before it’s too late and there’s nothing more to save. “Why did the Apostrophe Protection Society not have a militant wing? Could I start one?”, she asks in the “Introduction”. And I must admit I sympathized with her feelings fully while reading the book for the first time, and now as I re-read it I still sympathize with her.
The book has given me a lot of useful tips presented in a lively, easy-to-understand manner. I might disagree with the author in some cases (for example, when she attacks emoticons, rather unfairly from my point of view), but I’m very grateful to her for writing this book and to Rosemary for sending it to me. It has helped me to rediscover the English language and given me quite a few minutes of true joy. I highly recommend it, especially if you are often in doubt whether to write “it’s” or “its”. Lynne explains the difference in such a way that you will never forget.

I almost fell over laughing, as a matter fact I was laughing, and he turned around and said; “what, you like my T-shirt,” in a New Jersey accent. Indeed, I said “I sure do, where to get it?” He told me he had it made because his brother-in-law owns a T-shirt company back in Jersey. That’s just too funny, and I asked him what you have against religious folks? He said; “why, are you religious,” and I said; “hell no” and he basically explained to me that he didn’t have anything against religious folks, as long as they keep that stuff to themselves.
I told him that it was interesting he was at a Chinese restaurant because most Chinese agree with him, as most of them are atheists. Then I asked him what he had against socialists, and he said; “why did you vote for Obama or what?” Again in a harsh New Jersey accent. And once again, I said; “hell no!” And then he said well you must have at least some intelligence, but is it over 140 IQ? Indeed, I told him; “well, I’m not lost yet,” and he smiled.
We both picked up our meals got in our perspective vehicles and took off, but I thought it was refreshing that the gentleman wore that T-shirt out in public, was obviously not politically correct, but didn’t give a rat’s rear end. He wasn’t about to take any crap from anyone, and apparently he’s is tired as many of us are out here in the real world with all that nonsense which goes on in our society. I would have stayed and chatted longer, but our food was ready. It might be interesting to find out what he thinks about other things as well. But what do you think?
]]>When you use a conventional publisher for your book they are taking a significant gamble with their own money. In their terms they want to see a return on that investment, some thousands of dollars, and they use their internal operations, including editors and designers, to help make certain that the book will attract the greatest volume of sales.
So, as the publisher is risking his money and reputation, then their people will have a considerable control on the completed work, which may not be what the writer wanted. The author’s plans are then subjugated to the publishers desire to print what will sell.
As a self publisher then you take all the risk - but you can now take control and shape your own destiny. Your monetary risk can be limited, tens of dollars rather than tens of thousands; you are risking time and reputation, not cash.
Further, you will carry the can for the project as well. It is up to you to find, or act as, an editor, designer, project manager, manuscript preparer and so on. Many of these you can do yourself, but always ask yourself if that is the best way to get the result you are seeking.
Why self-publish anyway? If you are a serious, professional, writer (or aspiring to be one) whose books have mass appeal then go the traditional route, but for most of you the self publishing route is the one.. It is not always straightforward, there’s much to study as you progress, much of it can be readily discovered and you can get help from many sources.
Here are the three main stages when self publishing:
* Authoring the publication’s content
* Preparing the completed manuscript for printing and distribution including preparing the design and all contents, e.g. illustrations, Table of Contents, end matter, covers.
* Proposing a promotion and marketing plan and putting it into action.
If ever there was a dynamics of self publishing then this is it!
To begin, then is the writing. Because you are not going the traditional publisher route the content is entirely up to you. When you are writing for profit then you must pick a topic that people will be called to own, so do your research thoroughly. Only then write your book or manual in an engaging but straightforward way.
This is where it helps to have an editor. Your editor can be a friend who has a critical eye, can identify spelling mistakes, language errors and plot or information inconsistencies easily. Choose someone whose standing you respect. It’s well worth while getting another pair of eyes to check your book so that it will be the best it can be.
Remember, when writing you need to touch the heart of your readers. For technical or instructional writing then you do this through clarity of style, if you are writing a novel then it is the characters who come alive and the plot that drives the reader forwards to get that response. You are listening for the reader to say to themselves "I really like this, I want more".
